The study of Endothelin-1 effect on melanocyte migration in malignant melanoma

Abstract

|
Endothelin-1 is a 21 amino acid vasoconstrictor peptide originally. isolated from endothelial cells and keratinocytes. There are three isotype ET-1, ET-2 and ET-3. Only ET-1 involve human skin. Yada group indicated that human melanocyte possesed specific high affinity receptors for ET-1 and that ET-1 stimulated melanocyte proliferation and migration.
The purpose of this study was to determine weather endothelin-1 could enhance melanocyte migration or cell proliferation in malignant melanoma.
We performed Boyden Chamber Checkboard analysis by using control, 20nM, 40nM and 80nM ET-1 concentration in SK 28 cell line and SK30 cell line. The data were represented as percent of cell migration and statistical student¢¥s t-test was done.
The following results were obtained from this study;
1. In SK 28 cell line, the percentage of cell migration noted 100.0410.12% at control, 248.86 6.91% at 20nM, 265.249.73% at 40nM and 285.158.34 at 80nM.
ET-1 significantly (p<0.01) enhanced cell movement at the concentration of 20, 40, and 80 nM.
2. In SK 30 cell line, the percentage of cell migration noted 100.0410.12% at control, 282.86 8.26% at 20nM, 290.867.05% at 40nM and 263.864.27 at 80nM.
ET-1 significantly (p<0.01) enhanced cell movement at the concentration of 20,40, and 80 nM. Key Words : Endothelin-1, Malignant Melanoma.
